This compelling paper demonstrates that ceramide accumulates in the muscle of sub-groups of men at increased risk of type 2 diabetes. For instance, the offspring of type 2 diabetic patients display reduced ceramide degradation, while in obese patients there is increased de novo synthesis of ceramide. Interestingly, muscle ceramide concentration was found to be negatively correlated with insulin sensitivity. This study provides new evidence that muscle ceramide is higher among men at risk of type 2 diabetes.
